In this project we investigate the relationship between pulmonary inflammation and lung cancer using a multidisciplinary approach. Definition of reliable criteria for the assessment of individual risk of lung cancer in heavy smokers could improve future prevention strategies, diagnostic approaches and clinical management of this lethal disease and could influence changes in smoking habits. The recognition of biological, molecular and genetic risk factors of COPD and lung cancer may characterize the interplay between these two diseases. Information is gathered on pro-inflammatory mediators and cells in lung tissue by immunohistochemistry and on systemic inflammatory factors by proteomics approaches. Characterization of stromal cells phenotype and gene expression profile provides a framework for understanding the cross-talk between tissue microenvironment and transformed epithelial cells. Moreover, the setting of reliable animal model of lung cancers where testing tumor promotion by inflammation allows to study cells and molecules responsible of such effect as well as to prove, in mice, the virtual risk factors emerging from human studies.
